AlphaScreen®, developed by Perkin Elmer, is a bead-based, proximity assay technology which requires a high-energy excitation at 680 nm and reading emission at 520-620 nm.
Three AlphaScreen® cartridges are available for the Paradigm Platform. The Standard cartridge is ideal for occasional AlphaScreen® users. The HTS cartridge is optimized for high-sensitivity and high-throughput 96- and 384-well screens and the 1536 cartridge is optimized for the smaller optical geometry of high-density plates, avoiding cross-talk from adjacent wells.

Molecular Devices CatchPoint® Assay Kits measure cAMP or cGMP levels via a competitive immunoassay format, similar to an ELISA. The assays' high affinity reagents are optimized for sensitivity and precision in applications where cAMP or cGMP levels are low. CatchPoint assays are quantitative, allowing users to accurately determine the amount of cAMP or cGMP in samples with rapid development (less than 10 minutes) and no stop step.
CatchPoint® assays are fast and simple using the TUNE optical cartridge with Paradigm Platform. Using the TUNE cartridge, dedicated excitation and emission filters are not required; the appropriate wavelengths are simply selected.
Figure 1: cAMP calibration curve. The calibration curve for the CatchPoint® cAMP Assay Kit was generated on a SpectraMax® M5 Multi-Mode Microplate Reader. Data were taken 24 hours after addition of Stoplight Red substrate. The EC50 of the cAMP calibration curve was 5.1 nM.

Bioluminescence Resonance Energy Transfer (BRET) is similar to FRET in that it utilizes a non-radiative energy transfer between a donor and an acceptor but rather than a fluorescent donor molecule a luminescent donor molecule is used.

Designed specifically for use with Perkin Elmer's BRET2 reagents, Paradigm Platform's Dual Luminescence Detection cartridge utilizes a patent-pending well-isolating design that significantly reduces the level of cross talk from surrounding wells. The resulting performance is ideal for BRET2 applications. Paradigm Platform's TUNE cartridge supports fluorescence intensity applications at any excitation wavelength between 360 and 790 nm and any emission wavelength between 400 and 840 nm, with sensitivity up to 10x higher than other monochromator optical systems. Cartridges with specific excitation and emission filter sets are available that occupy a single bay, leaving room for additional cartridges, such as Absorbance or AlphaScreen, to be installed at the same time (three top and three bottom read positions are available).

Molecular Devices University on Microplate Assays: DNA, Protein Analysis and Immunodetection
Fluorescence Polarization (FP) is a homogenous assay format that relies on monitoring molecular movement and rotation. Most FP assays employ a small fluorescently-labeled molecule that binds to a larger molecule, causing a decrease in molecular rotation and a change in fluorescence polarization. FP has been widely applied to monitoring enzymatic assays.
FP requires monitoring light through vertical and horizontal polarizing filters in addition to fluorescence band-pass filters. Paradigm Platform's dual PMT design enables simultaneous monitoring of both vertical and horizontal emissions, delivering much faster read times compared to a single PMT design. Since polarization filters attenuate light traveling through the optical path, any optical components that further degrade light transmission can affect performance. Paradigm Platform's direct excitation optical path eliminates photon-robbing components such as optical fibers.

HTRF®, developed by CisBio, combines the features of fluorescence resonance energy transfer (FRET) and TRF, using Europium (Eu) cryptate as its donor fluorophore and a modified allophycocyanin (XL665) or the small molecule d2 as the acceptor fluorophore. When interacting biomolecules are labeled with Europium cryptate and d2, FRET takes place between the donor and the acceptor fluorophores.
HTRF® assays with Paradigm Platform are fast to run and simple to analyze. The HTRF® Detection cartridge features simultaneous emission detection, rather than sequential detection, of the donor and acceptor, yielding faster and more accurate results. Data acquisition and analysis are simple with ready-to-use protocols in the integrated SoftMax Pro software. Well-to-well variability is automatically corrected using the patented HTRF® ratio calculation (used under license). Other commonly used data reduction tools including Delta Ratio and Delta F% are also available.
